研究領(lǐng)域和方向
針對(duì)臨床未滿足的重大疾病需求,如超級(jí)耐藥菌感染、關(guān)節(jié)炎、胰腺炎、敗血癥、抑郁癥、腦部外創(chuàng)傷、老年癡呆癥等,從具有成藥性優(yōu)勢(shì)的天然骨架(如大環(huán)內(nèi)酯、小檗堿、血根堿、紫檀芪、甘草次酸、碳苷黃酮等)出發(fā),利用現(xiàn)代化學(xué)生物學(xué)、蛋白質(zhì)組學(xué)、結(jié)構(gòu)生物學(xué)以及人工智能計(jì)算機(jī)尋靶等手段尋找新的靶標(biāo)或者作用模式,合成生物學(xué)或有機(jī)化學(xué)合成新分子,突破現(xiàn)有的治療手段的局限性。通過(guò)迭代藥物設(shè)計(jì)和改造結(jié)構(gòu),提高提高先導(dǎo)化合物的生物活性和類藥性,同時(shí),在化學(xué)和生物學(xué)的交叉層面中闡明分子作用機(jī)制,最終獲得針對(duì)重大疾病模型的具有體外、體內(nèi)活性的臨床候選化合物。
